Make sure your firewall and/or antivirus isn't blocking it.

Also, if you're on Vista, make sure to run as Admin.

Turn on mic boost.

Right click on the sound icon in the tray, and go to properties.  Switch over to recording, and in one of the menus there should be mic boost.

If you can't find it, there's many guides on the internet.

Go into Sound Recorder (In Accessories somewhere), and try recording yourself and playing it back.

This usually happens to me as well. However, in order to fix it, all I would do is go into my control panel, then sounds and audio devices.

From there, click on the Voice tab, and there should be a button to "Test Hardware." Upon completing the test, it usually will recalibrate things and your mic should be functional in TS. Before running the "test hardware" feature, I would suggest you completely close out of TS or Vent prior to using it.
